The seed of the hemp plant (Cannabis sativa L.) has been revered as a nutritional resource in Old World Cultures. This has been confirmed by contemporary science wherein hempseed oil (HSO) was found to exhibit a desirable ratio of omega-6 and omega-3 polyunsaturated fatty acids (PUFAs) considered optimal for human nutrition. HSO also contains gamma-linoleic acid (GLA) and non-psychoactive cannabinoids, which further contribute to its’ potential bioactive properties. Herein, we present the kinetics of the thermal stability of these nutraceutical compounds in HSO, in the presence of various antioxidants (e.g. butylated hydroxytoluene, alpha-tocopherol, and ascorbyl palmitate). We focussed on oxidative changes in fatty acid profile and acidic cannabinoid stability when HSO was heated at different temperatures (25 °C to 85 °C) for upto 24 h. The fatty acid composition was evaluated using both GC/MS and 1H-NMR, and the cannabinoids profile of HSO was obtained using both HPLC-UV and HPLC/MS methods. The predicted half-life (DT50) for omega-6 and omega-3 PUFAs in HSO at 25 °C was about 3 and 5 days, respectively; while that at 85 °C was about 7 and 5 hours respectively, with respective activation energies (Ea) being 54.78 ± 2.36 and 45.02 ± 2.87 kJ/mol. The seeds of hemp (Cannabis sativa L.) have been an important source of nutrition for thousands of years in Chinese and European cultures but, nowadays, in spite of the multiple clinical evidence, which highlights their huge health-promoting properties, people are still unaware about their nutritional as well as nutraceutical benefits. Indeed, the extraordinary richness of essential polyunsaturated fatty acids is odds with oxidative stability of hemp seed by-products. This is the reason for which different and innovative extractive strategies are daily employed, as well as the preservation of antioxidant hemp seed phenols and polyphenols is claimed as a key issue to pursue. Recent insights into the phenol and cannabinoid baggage of hemp seed highlight it is an inestimable source of health promoting ingredients. This review aims to provide a detailed and updated background for research on seeds of this niche crop, pointing out the chemistry and bioactivity of their phytochemicals.

Multiple Sclerosis (MS), is a common autoimmune disease that affects the central nervous system. Currently, there is no cure for MS, and most treatments involve the use of immunosuppressive drugs that can have adverse effects or increased toxicity. Cannabis, commonly known as marijuana, is a product of the Cannabis sativa , and for several centuries has been used as an alternative medicine in many cultures. Cannabis sativa produces over 421 chemical compounds, including about 80 terpenophenols named phytocannabinoids, and include both psychotropic THC and non-psychoactive CBD. In the currently study, we show that a combination therapy using THC and CBD results in amelioration of EAE, an animal model of MS, by reducing hind limb paralysis, decreasing immune cellular infiltration into the brain, and mitigating the presence of inflammatory biomarkers, including gram negative bacteria-associated lipopolysaccharide (LPS). Interestingly, the gut microbiome plays an important role in immune function and studies have shown that it is altered significantly in MS patients. Inasmuch, we performed 16S rRNA sequencing on experimental groups to investigate the gut microbiome composition after using a combination of THC and CBD compared to disease controls. Interestingly, we found that EAE mice showed increase in the mucin degrading bacterial species, Akkermansia municiphila , which was significantly reduced in disease mice treated with THC+CBD. Collectively, our data suggests that THC+CBD can ameliorate EAE by preventing accumulation of mucin-degrading bacteria that would lead to increased gut microbial dysbiosis.

Cannabidiol (CBD), a major constituent of Cannabis, has been shown to be a powerful anti-inflammatory and anti-anxiety drug, without exerting a psychotropic effect. However, when given either intraperitoneally or orally as a purified product, a bell-shaped dose-response was observed, which limits its clinical use. In the present study, we have studied in mice the anti-inflammatory and anti-nociceptive activities of standardized plant extracts derived from the Cannabis sativa L., clone 202, which is highly enriched in CBD and hardly contains any psychoactive ingredients. In stark contrast to purified CBD, the clone 202 extract, when given either intraperitoneally or orally, provided a clear correlation between the anti-inflammatory and anti-nociceptive responses and the dose, with increasing responses upon increasing doses, which makes this plant medicine ideal for clinical uses. The clone 202 extract reduced zymosan-induced paw swelling and pain in mice, and prevented TNFα production in vivo. It is likely that other components in the extract synergize with CBD to achieve the desired anti-inflammatory action that may contribute to overcoming the bell-shaped dose-response of purified CBD. We therefore propose that Cannabis clone 202 (Avidekel) extract is superior over CBD for the treatment of inflammatory conditions.
