What Is Glutathione? Reduced glutathione, commonly known as glutathione or GSH, is a tripeptide consisting of L-glutamine, L-cysteine, and glycine. It is ubiquitous in living systems. Glutathione bio-synthesis can be affected by biochemical individuality and/or dietary factors. Chronic oxidative stress can also deplete cellular glutathione. Glutathione is naturally synthesized from N-Acetyl Cysteine (NAC) in the human body. Precursors to glutathione, such as whey protein, vitamin C, and glutamine, are also recommended to boost glutathione levels in the body; however, results are inconsistent. Biological individuality is such that not every body has equivalent ability to metabolize the precursor to raise glutathione.* Why Not Give Pure Glutathione? Unfortunately, most oral forms of glutathione are foul smelling, but more importantly, the majority of an oral dose is oxidized before it can be absorbed and used by the cells. Glutathione (GSH) plays an important role in a multitude of cellular processes, including cell differentiation, proliferation, and apoptosis, and as a result, disturbances in GSH homeostasis are implicated in the etiology and/or progression of a number of human diseases, including cancer, diseases of aging, cystic fibrosis, and cardiovascular, inflammatory, immune, metabolic, and neurodegenerative diseases. Owing to the pleiotropic effects of GSH on cell functions, it has been quite difficult to define the role of GSH in the onset and/or the expression of human diseases, although significant progress is being made. GSH levels, turnover rates, and/or oxidation state can be compromised by inherited or acquired defects in the enzymes, transporters, signaling molecules, or transcription factors that are involved in its homeostasis, or from exposure to reactive chemicals or metabolic intermediates. GSH deficiency or a decrease in the GSH/glutathione disulfide ratio manifests itself largely through an increased susceptibility to oxidative stress, and the resulting damage is thought to be involved in diseases, such as cancer, Parkinson's disease, and Alzheimer's disease. In addition, imbalances in GSH levels affect immune system function, and are thought to play a role in the aging process. Just as low intracellular GSH levels decrease cellular antioxidant capacity, elevated GSH levels generally increase antioxidant capacity and resistance to oxidative stress, and this is observed in many cancer cells. The higher GSH levels in some tumor cells are also typically associated with higher levels of GSH-related enzymes and transporters. Although neither the mechanism nor the implications of these changes are well defined, the high GSH content makes cancer cells chemoresistant, which is a major factor that limits drug treatment. The present report highlights and integrates the growing connections between imbalances in GSH homeostasis and a multitude of human diseases.

Fucoidan is a sulfated polysaccharide widely distributed in brown seaweed. It exhibits several bioactivities, such as anti-cancer, anti-tumor, anti-microbial, anti-diabetic and anti-oxidant properties. However, the effects of fucoidan in chondrocytes are not well established. Previously, we have reported in vitro and in vivo anti-inflammatory effects of fucoidan. In this study, we evaluated the effects and regulatory mechanism of fucoidan derived from Undaria pinnatifida on the cyclooxygenase-2 (COX-2) and type II collagen in rabbit articular chondrocytes. Using western blotting and alcian blue staining, respectively, fucoidan was shown to induce type II collagen and sulfated proteoglycan in a dose- and time-dependent manner. Moreover, fucoidan inhibited the COX-2 expression in a dose- and time-dependent manner and increased the phosphorylation of extracellular signal-regulated kinase (ERK), p38, and AKT kinases in chondrocytes. The inhibition of p38 and AKT using SB203580 and LY294002, respectively, in the presence of fucoidan decreased the expression of type II collagen. However, ERK inhibition using PD98050 stimulated type II collagen expression. Fucoidan increased COX-2 expression in the presence of inhibitors of ERK, p38, and AKT kinases. These results conclusively suggested that fucoidan regulated type II collagen expression via the p38 and AKT pathways, and COX-2 expression via the p38, ERK and AKT pathways in rabbit articular chondrocytes. Moreover, given its ability to mediate cell differentiation and exert anti-inflammatory activity, fucoidan may represent a potential therapeutic substance for use in inflammatory conditions, including arthritis.

If we're talking about a preventative dose, meaning you're not sick, then anywhere between 1200 and 2000 milligrams per day. It was this quantity that was given to elderly patients that showed results? So that 1200 milligram a day mark showed a reduced severity of symptoms, reduced length of time of illness and reduction in the recurrence of illness. So 1200 to 2000 milligrams a day is where you are looking as your preventative dose. Now kind of a more aggressive moderate dose if you think you're coming down with something would be somewhere in between 2000 and 4,000 milligrams per day. And if you're full blown in bed with super illness - if you were in the hospital (and it was a nutritionally oriented hospital), they would actually potentially IV you with N-Acetylcysteine to the order of magnitude of a hundred milligrams per kilogram a day, which equates to about eight grams or 8,000 milligrams, which is 2000 more than what some viruses are shown to deplete from the body. So it gives you the 6,000 plus an additional 2000, depending on your weight again, but that's for the average 150 pound person. If you're super sick and you're trying to reap the potential benefit of what research has shown with NAC, these are levels that you could consider taking. Now for some people this higher dose, what may happen is nausea or upset stomach might be the limiting factor and prevent you from being able to get that much. Some people say "let's take it to tolerance". Let's take it up to this level or to our ability to tolerate it. So maybe you fall shy at 5,000, your stomach is really upset. You can't handle any more. That's okay. You're still getting some of that in and you're still working yourself toward the potential of protecting yourself. So again, NAC is one of the most powerful and one of the most effective ways to shorten the duration of a viral infection, but also to help reduce the symptom severity and to reduce the risk of a recurring infection.
