The Effect of Aloe Vera Solution on Chemotherapy-Induced Stomatitis in (...)
Background: Stomatitis is the most common complication of chemotherapy. This study aimed to assess the effect of aloe vera solution on stomatitis and its pain intensity in patients undergoing chemotherapeutic procedures.Methods: In this randomized controlled clinical trial, 64 patients with Acute Myeloid Leukemia and Acute Lymphocytic Leukemia undergoing chemotherapy were randomly divided into a control and an intervention group. The intervention group patients were asked to wash their mouths with 5 ml of aloe vera solution for two minutes three times a day for 14 days. The control group patients, however, used only the ordinary mouthwashes recommended in hematologic centers. The patients’ mouths were examined by two assistants on days 1, 3, 5, 7, and 14. The intensity of stomatitis was recorded according to WHO stomatitis intensity checklists and pain was evaluated using Visual Analog Scale. The data were analyzed by SPSS statistical software, version 18.Results: The results showed that aloe vera solution mouthwash significantly reduced the intensity of stomatitis and its pain in the intervention group compared to the control group. On the first day, no significant difference was found between the two groups regarding the mean intensity of stomatitis (P=0.178) and pain (P=0.154). However, a significant difference was observed between the two groups in this regard on other days (days 3-14: P=0.001 for stomatitis intensity, P=0.001 for pain).Conclusions: Aloe vera solution can improve the patients’ nutritional status, reduce stomatitis and its pain intensity, and increase the patients’ satisfaction.Trial Registration Number: IRCT2014092819318N1

Background: Diabetes mellitus is a disorder in the metabolism of sugars, fats and proteins in the body, leading to hyperglycemia and hyperlipidemia. Resistin is an adipocytokine associated with diabetes. This study aimed at investigating the effect of high intensity interval training and Aloe Vera consumption on resistin and the insulin resistance index in diabetic rats. Materials and Methods: In this experimental study, 32 male Wistar streptozotocin-(STZ) diabetic rats were randomly divided into four groups including the control, training, supplementation and training + supplementation. The training program included 8 weeks, five sessions per week, and each session with 6-12 periods of two minutes activity at 90 maximum speed and one minute rest (10 m/min). The Aloe Vera gel (300 mg/kg) was given as gavage in the supplementation recipient groups, 5 days a week for 8 weeks. Results: The results showed that the insulin levels and insulin resistance index increased and resistin decreased significantly in the experimental groups (P0.05). Conclusion: It seems that combination of high intensity interval training and the use of Aloe Vera can have a positive effect on diabetes control by reducing resistin and glucose and increasing insulin levels.

Introduction : Tuberculosis has become a serious health  problem in the world. Increased mortality in patients with tuberculosis infection is associated with MDR and hepatotoxic side effects of antituberculosis drugs. Therefore, it is necessary to find an alternative drug that can reduce the hepatotoxic side effects . Objective : This study aims to investigate the benefits of Aloe vera in preventing liver damage as a side effect of antituberculosis drug administration. Method: Twenty-five rats will be divided into five groups, namely the control group (without any treatment), the group of rats treated by anti-tuberculosis drugs, and a group of mice were given antituberculosis drugs and got Aloe vera extract at a dose of 40; 80; and 120 mg/kg body weight. Results: Antituberculosis drugs are isoniazid and rifampicyn a dose of 50 mg/kg body weight. Levels of TNF-a were significantly higher in the antituberculosis group compared with the control group (P < 0.05). All doses of Alovevera reduce levels of TNF-a compared with the antituberculosis group (P < 0.05), although it has not yet reached levels comparable to the control group (P > 0.05). Conclusion: It was concluded that administration of Aloe vera can suppress the production of TNF-a as a result of antituberculosis drug administration. Thus, Aloe vera can be a useful alternative to natural materials in the successful treatment of tuberculosis through the inhibition of hepatotoxid side effect.   Keywords: Aloe vera; TNF-a; antituberculosis drugs.

Background: Thyroid cancer has been growing rapidly during the last decades. Radioiodine-131 (I-131) as an appropriate therapy modality is currently using in the treatment of cancer and hyperthyroidism diseases. This radiotracer is considered as a cause of oxidative DNA damage in nontarget cells and tissues. The aim of this study was to investigate the effects of curcumin and trehalose on the level of DNA double-strand breaks (DSBs) caused by I-131 in human lymphocytes. Materials and Methods: First, 6-mL blood samples were taken from each of the five volunteers. After 1 h of preincubation with the antioxidants, a total of 20 μCi I-131/2 mL (blood + NaCl) was added to each sample, and then, the samples were reincubated for 1 h. Lymphocytes were separated and the mean DSB levels were measured for each sample through γ-H2AX assay to evaluate the effects of antioxidants. Results: After 1-h incubation with I-131, the DSBs increased by 102.9% compared to the control group (0.343 vs. 0.169 DSB/cell; P = 0.00). Furthermore, compared to the control + I-131 group, curcumin and trehalose reduced the DSBs by 42% and 38%, respectively. There was a significant decrement (P = 0.00) in the levels of DSBs of the curcumin + I-131 and trehalose + I-131 subgroups compared to the control + I-131 subgroup. Furthermore, there was no significant relationship between the radioprotective effect of curcumin and trehalose (P = 0.95). Conclusion: The use of curcumin and trehalose as antioxidant can reduce the numbers of DSBs caused by I-131. Meanwhile, the radioprotective effect of curcumin was more than trehalose.

Trehalose is widely used as a sweetener, humectant, and stabilizer, but is ubiquitously degraded by the enzyme trehalase expressed in a broad variety of organisms. The stability of the new trehalose analogues lentztrehaloses A, B, and C in microbial and mammalian cell cultures and their pharmacokinetics in mice were analyzed to evaluate their potential as successors of trehalose. Among the 12 species of microbes and 2 cancer cell lines tested, 7 digested trehalose, whereas no definitive digestion of the lentztrehaloses was observed in any of them. When orally administered to mice (0.5 g/kg), trehalose was not clearly detected in blood and urine and only slightly detected in feces. However, lentztrehaloses were detected in blood at >1 μg/mL over several hours and were eventually excreted in feces and urine. These results indicate that lentztrehaloses may potentially replace trehalose as nonperishable materials and drug candidates with better bioavailabilities.

Marine Drugs Free Full-Text Fucoidan Structure and Activity in Relation to Anti-Cancer Mechanisms
Fucoidan is a natural derived compound found in different species of brown algae and in some animals, that has gained attention for its anticancer properties. However, the exact mechanism of action is currently unknown. Therefore, this review will address fucoidans structure, the bioavailability, and all known different pathways affected by fucoidan, in order to formulate fucoidans structure and activity in relation to its anti-cancer mechanisms. The general bioactivity of fucoidan is difficult to establish due to factors like species-related structural diversity, growth conditions, and the extraction method. The main pathways influenced by fucoidan are the PI3K/AKT, the MAPK pathway, and the caspase pathway. PTEN seems to be important in the fucoidan-mediated effect on the AKT pathway. Furthermore, the interaction with VEGF, BMP, TGF-&beta;, and estrogen receptors are discussed. Also, fucoidan as an adjunct seems to have beneficial effects, for both the enhanced effectiveness of chemotherapy and reduced toxicity in healthy cells. In conclusion, the multipotent character of fucoidan is promising in future anti-cancer treatment. However, there is a need for more specified studies of the structure&ndash;activity relationship of fucoidan from the most promising seaweed species.

Fucoidan is a sulfated polysaccharide widely distributed in brown seaweed. It exhibits several bioactivities, such as anti-cancer, anti-tumor, anti-microbial, anti-diabetic and anti-oxidant properties. However, the effects of fucoidan in chondrocytes are not well established. Previously, we have reported in vitro and in vivo anti-inflammatory effects of fucoidan. In this study, we evaluated the effects and regulatory mechanism of fucoidan derived from Undaria pinnatifida on the cyclooxygenase-2 (COX-2) and type II collagen in rabbit articular chondrocytes. Using western blotting and alcian blue staining, respectively, fucoidan was shown to induce type II collagen and sulfated proteoglycan in a dose- and time-dependent manner. Moreover, fucoidan inhibited the COX-2 expression in a dose- and time-dependent manner and increased the phosphorylation of extracellular signal-regulated kinase (ERK), p38, and AKT kinases in chondrocytes. The inhibition of p38 and AKT using SB203580 and LY294002, respectively, in the presence of fucoidan decreased the expression of type II collagen. However, ERK inhibition using PD98050 stimulated type II collagen expression. Fucoidan increased COX-2 expression in the presence of inhibitors of ERK, p38, and AKT kinases. These results conclusively suggested that fucoidan regulated type II collagen expression via the p38 and AKT pathways, and COX-2 expression via the p38, ERK and AKT pathways in rabbit articular chondrocytes. Moreover, given its ability to mediate cell differentiation and exert anti-inflammatory activity, fucoidan may represent a potential therapeutic substance for use in inflammatory conditions, including arthritis.
