The use of fucoidan, a marine-origin bioactive polymer, is herein proposed as a component of an innovative and effective strategy against melanoma, one of the most aggressive skin cancers. First, fucoidan antitumor activity, in its soluble form, was assessed presenting increased cytotoxicity over melanoma cells when compared to human dermal fibroblasts and keratinocytes. After this antitumor activity validation and trying to develop a more targeted and local strategy aiming to diminish the cytotoxic effects over noncancer cells, fucoidan was immobilized at the surface of an electrospun nanofiber mesh (NFM_Fu), envisioning the development of a therapeutic patch. The maximum immobilization concentration was 1.2 mg mL–1, determined by the Toluidine Blue Assay and confirmed by XPS. Furthermore, NFM_Fu is more hydrophilic than NFM, presenting a contact angle of 36°, lower than the 121° of the control condition. NFM_Fu was able to reduce human melanoma cell viability by 50% without affecting human dermal fibroblasts and keratinocytes. Taken together, these results set the basis for a valuable approach for melanoma treatment.

Hydroactive wound dressings ensure a physiologically moist wound milieu which enhances healing and patient comfort. Polymers like polyurethane, salts of alginic acid, collagen, cellulose and other gelable polysaccharides are commonly used for preparation of hydrogels but have their own limitations. In the present study acacia gum, a natural polysaccharide having known antioxidant and wound healing properties has been blended with polyvinyl alcohol and prepared hydrogel matrix further explored for its wound healing potential. Structural characterization of blended films by FTIR and XRD method showed that polysaccharide gum associated with PVA molecules via acetal bridges and they were amorphous in nature. The gel exhibited free swell capacity of 64 g 0.100 cm−2, which is comparable to the commercially available films recommended for heavily exuding wounds. The GA/PVA blends showed 34% moisture retention (Rh) ability after 24 h and their fluid absorbing (26%) and fluid donation (16%) ability make them suitable for moist and fibrinous wounds. They also exhibited blood compatibility, oxygen permeability, bacterial impermeability, antioxidant activity, iron chelation ability and biodegradability. In vivo healing potential has been evaluated on Swiss albino mice where hydrogel coated wounds showed faster and scar-less wound regeneration. In vitro Povidone-Iodine loading and release studies showed that iodine release followed Fickian diffusion process. Prepared hydrogels are breathable and hydrophilic in nature and are able to maintain appropriate moisture level at wound surface, suitable for acceleration of wound healing process. Based upon the studied properties the designed hydrogels are recommended for dry, necrotic and low exuding wounds.

Microorganisms Free Full-Text Role of L. plantarum KX519413 as Probiotic and Acacia Gum as Prebiotic in Gastrointestinal Tract Strengthening
Probiotics, prebiotics, and synbiotics are potential mediators to maintaining healthy intestinal flora and have garnered an area of wide research in the past few years. The current study assesses the in vivo effects of probiotic (Lactobacillus plantarum MBTU-HK1), prebiotic (acacia gum) (either singly or in combination as a synbiotic on growth performance), biochemical, hematological, physiological, and immunological effects and their role in the reduction of procarcinogen enzyme activities in male Balb/c mice. The absence of treatment-related toxicity and a normal physiological range of biochemical and hematological parameters ensure their safe consumption. The synbiotic group was found to possess lowered cholesterol levels and enhanced protein and mineral content. The probiotic and synbiotic groups reinforced immunoglobulin levels and had a modulatory effect on phagocytosis. A lymphocyte proliferation pattern suggested the stimulatory effect of synbiotic combination on splenocyte viability and proliferation. Total antioxidant capability in the liver was determined by a 2,2-diphenyl-1-picrylhydrazyl assay and all the treatment groups were found to possess increased scavenging activity. Synbiotic and prebiotic treatment was observed to lead to reduced tumor necrosis factor α (TNF-α) levels. Bacterial procarcinogenic fecal enzyme activities were found to be decreased, proving their role in the prevention of colon cancer incidence. This study proves the potency and safety of oral administration of L. plantarum MBTU-HK1 and acacia gum either individually or in combination.
