Marine Drugs Free Full-Text Fucoidan from Undaria pinnatifida Ameliorates Epidermal Barrier Disruption via Keratinocyte Differentiation and CaSR Level Regulation HTML
The epidermal barrier acts as a line of defense against external agents as well as helps to maintain body homeostasis. The calcium concentration gradient across the epidermal barrier is closely related to the proliferation and differentiation of keratinocytes (KCs), and the regulation of these two processes is the key to the repair of epidermal barrier disruption. In the present study, we found that fucoidan from Undaria pinnatifida (UPF) could promote the repair of epidermal barrier disruption in mice. The mechanistic study demonstrated that UPF could promote HaCaT cell differentiation under low calcium condition by up-regulating the expression of calcium-sensing receptor (CaSR), which could then lead to the activation of the Catenin/PLCγ1 pathway. Further, UPF could increase the expression of CaSR through activate the ERK and p38 pathway. These findings reveal the molecular mechanism of UPF in the repair of the epidermal barrier and provide a basis for the development of UPF into an agent for the repair of epidermal barrier repair.

Background: Pressure ulcers have afflicted critically ill patients since the dawn of recorded medicine. Pressure ulcers had adverse patient outcomes. Aloe vera as a non-pharmacological modality can be used in the pressure ulcer management. Aim of the study: to investigate the effect of Aloe Vera on the healing of second degree pressure ulcers among critically ill patients. Research design: A quasi-experimental research design was used to conduct this study. Sample: 60 adult critically ill patients developed second-degree pressure ulcer. Setting: Intensive Care Units of Emergency Hospital Mansoura University. Results: There were highly statistical significant differences between study and control groups regarding pressure ulcer wound's characteristics: size, depth, edges, wound undermining presence, skin color surrounding wound, granulation tissues presence, and epithelial tissues presence (P < 0.001**) at post intervention assessment. Conclusion: Aloe vera cream is effective in management pressure ulcers as it stimulates and accelerates the healing process. It is excellent, cheaper and safer choice than current conventional therapeutic management of pressure ulcers. Therefore, it can be routinely employed in management of pressure ulcers. Recommendation: Encourage the establishment of non-pharmacological remedies using (aloe vera) in treatment of pressure ulcer wounds and other different types of wounds.

Background: Diabetes mellitus is a disorder in the metabolism of sugars, fats and proteins in the body, leading to hyperglycemia and hyperlipidemia. Resistin is an adipocytokine associated with diabetes. This study aimed at investigating the effect of high intensity interval training and Aloe Vera consumption on resistin and the insulin resistance index in diabetic rats. Materials and Methods: In this experimental study, 32 male Wistar streptozotocin-(STZ) diabetic rats were randomly divided into four groups including the control, training, supplementation and training + supplementation. The training program included 8 weeks, five sessions per week, and each session with 6-12 periods of two minutes activity at 90 maximum speed and one minute rest (10 m/min). The Aloe Vera gel (300 mg/kg) was given as gavage in the supplementation recipient groups, 5 days a week for 8 weeks. Results: The results showed that the insulin levels and insulin resistance index increased and resistin decreased significantly in the experimental groups (P0.05). Conclusion: It seems that combination of high intensity interval training and the use of Aloe Vera can have a positive effect on diabetes control by reducing resistin and glucose and increasing insulin levels.

Protein aggregation has been proved to be a pathological basis accounting for neuronal death caused by either transient global ischemia or oxygen glucose deprivation (OGD), and inhibition of protein aggregation is emerging as a potential strategy of preventing brain damage. Trehalose was found to inhibit protein aggregation caused by neurodegenerative diseases via induction of autophagy, whereas its effect is still elusive on ischemia-induced protein aggregation. In this study, we investigated this issue by using rat model of transient global ischemia and SH-SY5Y model of OGD. We found that pretreatment with trehalose inhibited transient global ischemia-induced neuronal death in the hippocampus CA1 neurons and OGD-induced death in SH-SY5Y cells, which was associated with inhibition of the formation of ubiquitin-labeled protein aggregates and preservation of proteasome activity. In vitro study showed that the protection of trehalose against OGD-induced cell death and protein aggregation in SH-SY5Y cells was reversed when proteasome activity was inhibited by MG-132. Further studies revealed that trehalose prevented OGD-induced reduction of proteasome activity via suppression of both oxidative stress and endoplasmic reticulum stress. Particularly, our results showed that trehalose inhibited OGD-induced autophagy. Therefore, we demonstrated that proteasome dysfunction contributed to protein aggregation caused by ischemic insults and trehalose prevented protein aggregation via preservation of proteasome activity, not via induction of autophagy.

Effects of Laminaria Japonica Polysaccharides on the Survival of Non-Small-Cell Lung Cancer A549 Cells
Objective. To investigate the effect of Laminaria japonica polysaccharides (LJP) on the survival of non-small-cell lung cancer (NSCLC) A549 cells and its mechanism. Methods. In vitro: the cells were randomly divided into control group, LJP (5 mg/ml) group, LJP (10 mg/ml) group, and LJP (20 mg/ml) group. After corresponding treatment, the survival rate and the expression of proteins related to proliferation, apoptosis, epithelial-mesenchymal transition (EMT), and signaling pathway were detected by CCK8 assay and Western blot, respectively. In vivo: a xenograft model was established to detect the tumor volume and mass and the expression of the above pathway proteins. Results. Compared with the control group, LJP decreased the survival rate of A549 cells (), inhibited the protein expression of Ki67 and PCNA (), downregulated the expression of Bcl-2 while upregulated the expression of Bax, cl-caspase-3, and cl-caspase-9 (), upregulated the expression of E-cadherin, downregulated the expression of vascular endothelial growth factor (VEGF) and N-cadherin (), and downregulated β-catenin, transcription factor-4 (TCF4), and c-Myc protein expression levels (). In vivo: LJP decreased the volume and mass of the xenograft tumors and downregulated β-catenin, TCF4, and c-Myc protein expression levels compared with the control group (). Conclusion. LJP can inhibit the survival of non-small-cell lung cancer A549 cells in vitro, and its mechanism is related to the inhibition of activation of β-catenin/TCF4 pathway activation.

Background: Alcoholic liver disease is caused as a result of chronic alcohol consumption. In this study, we used an alcoholic liver injury mouse model to investigate the effect of fucoidan on ethanol-induced liver injury and steatosis and the underlying mechanisms. Methods: All mice were randomly divided into four groups: 1) control group, 2) model group, 3) diammonium glycyrrhizinate treatment group (200 mg/kg body weight), and 4) fucoidan treatment group (300 mg/kg body weight). Administration of ethanol for 8 weeks induced liver injury and steatosis in mice. Results: Fucoidan treatment decreased serum alanine aminotransferase activity, serum total cholesterol levels, and hepatic triglyceride levels, and improved the morphology of hepatic cells. Fucoidan treatment upregulated the expression of AMPKα1, SIRT1, and PGC-1α and inhibited the expression of ChREBP and HNF-1α. The levels of hepatic IL-6 and IL-18 were significantly decreased in the fucoidan group. Further, the levels of cytochrome P450-2E1 (CYP2E1), glucose-regulated protein (GRP) 78, and 3-nitrotyrosine (3-NT) in hepatic tissues were reduced in the fucoidan group as compared to the model group. Fucoidan significantly reversed the reduction of ileac Farnesoid X receptor (FXR) and fibroblast growth factor 15 (FGF15) levels induced by alcohol- feeding and reduced CYP7A1 (cholesterol 7α-hydroxylase) expression and total bile acid levels in the liver tissue. In addition, fucoidan regulated the structure of gut flora, with increased abundance of Prevotella and decreased abundance of Paraprevotella and Romboutsia as detected by 16S rDNA high-throughput sequencing. Conclusion: Fucoidan inhibited alcohol-induced steatosis and disorders of bile acid metabolism in mice through the AMPKα1/SIRT1 pathway and the gut microbiota–bile acid–liver axis and protected against alcohol- induced liver injury in vivo.

Abstract Biogenic synthesis of silver nanoparticles (AgNP) was performed at room temperature using Aloe vera plant extract in the presence of ammoniacal silver nitrate as a metal salt precursor. The formation of AgNP was monitored by UV-visible spectroscopy at different time intervals. The shape and size of the synthesized particle were visualized by scanning electron microscopy (SEM) and transmission electron microscopy (TEM) observations. These results were confirmed by X-ray powder diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR) analyses and further supported by surface-enhanced Raman spectroscopy/Raman scattering (SERS) study. UV-visible spectrum has shown a sharp peak at 420 nm and further evidenced by FTIR peak profile (at 1587.6, 1386.4, and 1076 cm−1 with corresponding compounds). The main band position with SERS was noticed at 1594 cm−1 (C–C stretching vibration). When samples were heated under microwave radiation, AgNP with octahedron shapes with 5–50 nm were found and this method can be one of the easier ways to synthesis anisotropic AgNP, in which the plant extract plays a vital role to regulate the size and shape of the nanoparticles. Enhanced antibacterial effects (two- to fourfold) were observed in the case of Aloe vera plant protected AgNP than the routinely synthesized antibiotic drugs.
